Isolation Of Enzyme Bromelin

The enzyme is known for the first time as a protein by Summer in 1926 that has managed to isolate urease from 'kara sword' (jack bean). Urease is an enzyme which can outline of urea to CO2 and NH3. Northop than few years and Kunitz to isolate pepsin, trypsin, chymotrypsin.
Furthermore, more and more enzymes that have been isolated and has been shown that this enzyme is a protein. Enzymes are chemical reactions or processes that take place well in the body which is possible because of the catalyst. The enzyme is a protease enzyme sulfidril bromelin of pineapple fruit. The presence of enzymes found in a state mixed with another protein that is required of a business separation to obtain pure enzyme. The separation is necessary to get special attention because the enzyme is highly dependent from the surrounding environment. On the separation of enzymes that need to be considered is the part where the enzyme is located, how the extraction, fractionation, purification, and purity test.

According to the IUB classification, enzyme bromelin have the code number EC 3.4.22.4 which means that the enzyme bromelin classified in group 3, ie EC hidrolase enzymes. The figures further show that successive bromelin classified protease enzyme, EC 3.4 enzyme bromelin means hydrolyze proteins / peptides. In IUB nomenclature, serine proteases, thiol proteases, metal proteases, and acid protease in a row have the EC number 3.4.21, EC 3.4.22, EC 3.4.23, and 3.4.24 EC number 4 (last digit) of the nomenclature of enzymes bromelin (EC 3.4.22.4) enzyme showed bromelin a thiol protease sequence to four in the list of serial enzyme. Thus the IUB nomenclature can at least take the information that bromelin enzyme is an enzyme catalyzing the hydrolysis reaction of proteins / peptides, which have active sisis containing thiol.

Variety Ananas comosus (pineapple) and some family Bromeliaceae, merupkan source proteolytic enzymes. Proteolytic activity of the pineapple plant has been known since the 19th century, especially in the trunk. Protease isolated from family Bromeliaceae is then called bromelin. The prospect of substantial development of this enzyme because this enzyme can be obtained from pineapple waste processing industries, such as liver, skin or pineapple tree. A large number of proteolytic enzymes which exist in the pineapple stem bromelin can not be known for certain. Bromelin crude can be separated by electrophoresis at pH 6.5 into four components that contain proteolytic activity. Ota et al. (1964) cited by Glazer and Smith (1977) succeeded in purifying bromelin from pineapple stems and fruit. Apparently there is no difference between the two sources of enzyme. However, enzymes from pineapple fruit bromelin contain fewer lissin, arginine, and histidine of the enzyme bromelin from pineapple stem. Titil stem bromelin enzyme is isoelectric at pH 9.5 was higher than in bromelin derived from pineapple fruit. In addition, bromelin not contain glucosamine, but has a total carbohydrate content of the larger.
